Neste trabalho confirmamos a resistência do feijão comum ( Phaseolus vulgaris) ao bruquídeo Callosobruchus maculatus (F.), inseto que preda sementes de feijão-de-corda (Vigna unguiculata) (L.). A resistência desta semente não está relacionada com a espessura do tegumento nem com os níveis de diversos compostos como ácido taníco, fenóis e ácido cianídrico conforme demonstram nossos resultados. No entanto, faseolina, detectada no tegumento por ‘‘Western blotting’’ e seqüenciamento N-terminal, é tóxica a C. maculatus. Esses dados estão de acordo com estudos anteriores feitos com duas outras sementes de leguminosas (Canavalia ensiformis e Phaseolus lunatus) e nos levam a sugerir que a presença de proteínas do tipo vicilina no tegumento de sementes de leguminosas tiveram papel importante nos mecanismo de adaptação de bruquídeos a sementes de leguminosas.

We have confirmed here that the seeds of the common bean (Phaseolus vulgaris, L.) do not support development of the bruchid Callosobruchus maculatus (F.), a pest of cowpea [Vigna unguiculata (L.) Walp] seeds. Analysis of the testa (seed coat) of the bean suggested that neither thickness nor the levels of compounds such as tannic acid, tannins, or HCN are important for the resistance. On the other hand, we have found that phaseolin (vicilin-like 7S storage globulin), detected in the testa by Western blotting and N-terminal amino acid sequencing, is detrimental to the development of C. maculatus. As for the case of other previously studied legume seeds (Canavalia ensiformis and Phaseolus lunatus) we suggest that the presence of vicilin-like proteins in the testa of P. vulgaris may have had a significant role in the evolutionary adaptation of bruchids to the seeds of leguminous plants.

A presença de insulina em plantas não é aceita pela comunidade científica em geral. Nesta revisão, discutimos esse paradigma e recuperamos informações que sugerem fortemente a presença de insulina em plantas. Adicionalmente, apresentamos resultados obtidos por nós revelando a presença em leguminosas de uma proteína com a mesma seqüência de aminoácidos da insulina bovina. Apresentamos, também, evidências da presença em plantas de proteínas normalmente associadas à sinalização por insulina em vertebrados.

The presence of insulin in plants is not accepted by the scientific community in general. In this review we discuss this paradigm and retrieve information that strongly suggests that insulin is indeed found in plants. We present results, which indicate that a protein molecule with the same amino acid sequence as bovine insulin is expressed in leguminous plants. Additionally, we provide evidence that proteins associated with insulin signalling pathways in vertebrates are also found in association with insulin-like molecules in plants.

Nesta revisão foram descritas definições, localizações, funções e exemplos de proteinases cisteínicas e suas proteinas inibidoras em animais vertebrados e invertebrados e plantas. Tratamos principalmente com aqueles inibidores que são relatados com o mecanismo de defesa da planta contra pestes. Em adição, comentamos sobre recentes trabalhos que contribuíram para uma melhor compreenção dos fatores envolvidos na interação específica proteinase cisteínica-cistatina. Por outro lado, chamamos atenção para o alto grau de afinidade e grande especificidade na interação que não são apenas representadas pela compatibilidade entre os residuos de aminoácidos do sítio ativo envolvidos na catalise, mas também de todos os resíduos de aminoácidos que participam da interação enzima-inibidor.

This review describeds the definition, localization, functions and examples of cysteine proteinases and their protein inhibitors in vertebrate, non-vertebrate animals and plants. These inhibitors are related with defense mechanisms of plant against pests. It also describes the factors involved in the specific cysteine proteinase-cystatin interaction and high degree of affinity and large specificity in this interaction which are not only represented by the compatibility between amino acid residues of the active site involved in catalysis, but also of all amino acid residues that participante in the enzyme-inhibitor interaction.

Vicilinas (globulinas de reserva 7S) isoladas de sementes de feijão-de-corda (Vigna unguiculata L.), susceptíveis (S) e resistentes (R) ao caruncho/gorgulho (Callosobruchus maculatus F., Coleoptera: Bruchidae) foram desnaturadas por uréia e fracionadas por cromatografia de troca iônica. As frações isoladas foram incorporadas em sementes artificiais para avaliação de sua toxicidade a C. maculatus. As fracões mais ácidas de ambas vicilinas afetaram o desenvolvimento e a sobrevivência do bruquídeo. Sugerimos que polipeptídeos de vicilinas de natureza tóxica são expressos em ambos tipos de globulinas de reserva, embora em níveis diferentes.

Vicilins (7S storage globulins) isolated from cowpea (Vigna unguiculata L.) seeds which were susceptible (S) and resistant (R) to the cowpea weevil (Callosobruchus maculatus F., Coleoptera: Bruchidae) were denatured by urea and fractionated by ion-exchange chromatography. Isolated fractions were incorporated in artificial seeds for assessment of their toxicity to C. maculatus. The most acidic fractions of both susceptible (CE-31 cultivar) and resistant (IT81D-1045 line) seeds were shown to affect development and survival of the bruchid. Results indicated that vicilin polypeptides of toxic nature were expressed in both types of storage globulins although at different levels.

Análise do látex de Manihot glaziovii mostrou a presença de várias atividades enzimáticas e inibitórias. O látex apresentou também um efeito inibitório sobre o desenvolvimento do gorgulho/caruncho do feijão-de-corda (Callosobruchus maculatus) em um sistema de sementes artificiais e sobre o desenvolvimento, em um ensaio in vitro, dos fungos fitopatogênicos Colletotrichum gloesporioides, Fusarium solani e Macrophomina phaseolina. Esses resultados sugerem a presença de substâncias, algumas delas de natureza protéica, envolvidas em mecanismos de defesa de plantas neste exsudato.

Analysis of the latex from Manihot glaziovii showed the presence of various enzymatic and inhibitory activities. The latex also presented an inhibitory effect on the development of cowpea weevil (Callosobruchus maculatus) in an artificial seed system and on the development, in an in vitro assay, of phytopathogenic fungi Colletotrichum gloesporioides, Fusarium solani and Macrophomina phaseolina. These results suggest the presence of substances, some of them of protein nature, involved in plant defense mechanisms in this exudation product.

O efeito da adição de quitina sobre o desenvolvimento e a sobrevivência do bruquídeo Callosobruchus maculatus (F.) foi estudado empregando-se um sistema de sementes artificiais feitas com farinha de Vigna unguiculata (feijão-de-corda). O polissacarídeo interferiu no desenvolvimento do inseto causando uma diminuição na taxa de sobrevivência. O índice de Howe mostra que as dietas contendo quitina são nutricionalmente inadequadas.
